Metabolism of Cyclic GMP in Vascular Smooth Muscle, Leukocytes, and Lung

Abstract

There are obvious historical reasons for the fact that our understanding of cyclic GMP metabolism in 1974 is far more limited than that relating to cyclic AMP. Over the almost 20 years that cyclic AMP has been known, the methodology has been consistently improved and refined whereas experience in the extraction, purification and measurement of cyclic GMP from tissues is much less extensive. The radioimmunoassay of Steiner et al. (1) with its sensitivity and specificity is obviously an extremely powerful tool. Nevertheless, perhaps in large part because of the low cyclic GMP content of most mammalian cells, there are often technical problems encountered in attempting to apply the assay successfully with tissue extracts (2, 3). In our laboratory it has been invariably necessary to purify cyclic GMP samples before assay, and to modify the purification procedures depending on the system studied.
