Abstract

Antagonistic roles for 5-phosphoribosyl 1-pyrophosphate (PPribose-P1) and purine nucleotides in effecting regulation of the rate of purine nucleotide synthesis de novo have been proposed from the kinetic and structural effects of these compounds on the allosteric enzyme PP-ribose-P amidotransferase described in the elegant studies of Holmes, Wyngaarden and Kelley (1, 2). From experiments in intact cells, Bagnara, Letter and Henderson (3) have pointed out additional or alternative mechanisms for the control of purine production such as the utilization of PP-ribose-P in competing reactions and purine nucleotide inhibition of PP-ribose-P synthesis.
