Chemical Modification and Cleavage of Proteins and Chemical Strategy in Immunochemical Studies of Proteins

Abstract

The determination of antigenic structures of proteins presents a major challenge, which is primarily chemical in nature. The chemical strategy set forth (Atassi, 1972) for our delineation of the complete antigenic structure of myoglobin (Atassi, 1975) has also proved extremely effective in substantial delineation of the antigenic structure of lysozyme. Delineation of the antigenic structures of these two proteins relied (Atassi, 1972) mainly on the following five approaches: (1) determination of the influence of conformational changes on immunochemical behavior, (2) study of the immunochemistry of specifically modified chemical derivatives of the protein, (3) study of the immunochemistry of a large number of peptide fragments with varying overlaps, (4) delineation of reactive regions on immunochemically reactive peptides by study of the immunochemistry of specific chemical derivatives of these peptides, and (5) organic synthesis of the antigenic reactive sites after they had been narrowed down by approaches (1)–(4) to a conveniently small size.
