Abstract

Three distinctive approaches are now available for the high-resolution, analytical fractionation of membrane proteins. These are: (1) Dodecylsulfate polyacrylamide gel electrophoresis (SDS-PAGE), where proteins are separated as SDS complexes according to molecular weight (Fairbanks et al., 1971; Weber and Osborn, 1969); (2) crossed immune electrophoresis (CIE), where the proteins, solubilized in nonionic detergent, are separated in one dimension according to charge and in a second dimension according to their antigenic properties (Bjerrum and Bøg-Hansen, 1976); and (3) Isoelectric focusing (IF), where the proteins, solubilized in nonionic detergent, are separated in a pH gradient, according to their isoelectric points.
