Evaluation of Known Genotoxic Agents and Δ9-Tetrahydrocannabinol for SCE Induction by the Intraperitoneal 5-Bromo-2’-Deoxyuridine Infusion Technique for Sister Chromatid Exchange Visualization

Abstract

A new technique for measurement of sister chromatid exchange (SCE) induction which utilizes intraperitoneal infusion of 5-bromo-2’-deoxyuridine (BrdU) for SCE visualization was evaluated using 7,12-dimethylbenz(a)anthracene (DMBA), a metabolically activated polyaromatic hydrocarbon with multiple development stage-specific biological effects, and methyl methane sulfonate (MMS), a direct-acting alkylating agent, in a transplacental system. Maternal bone marrow and whole fetus were examined. A recently developed procedure for isolating fetal cells was used which permitted, for the first time, the evaluation of SCE induction in fetal brain as well as other fetal organs.
