Abstract
Methyl methanesulfonate (MMS) and ethyl methanesulfonate (EMS) are highly mutagenic in lower organisms but only moderately carcinogenic in mice and rats. No case reports or epidemiological studies on the hazards of MMS and EMS to humans are known to the Working Group. This review summarizes and evaluates the data on genetic effects of these compounds from the aspect of their relevance to the assessment of possible genetic effects of these or related compounds in humans. First, a general summary is given of available data on MMS and EMS and some typical examples are given of dose-related effects of EMS and MMS in mice, Drosophila, and some other organisms. Then, molecular mechanisms of mutagenesis are discussed. Third, an attempt is made to assess the human hazard of these compounds on the basis of the experimental data and finally the areas requiring further investigation are considered.
