Summary
‘Prothrombin Complex’ is a mixture of a partially purified group of proteins (Factors II, VII, IX, and X) dependent upon vitamin K for biological activity. Treatment of congenital Factor IX deficiency is the best documented use for this mixture of proteins generally isolated from plasma or its subsequent fractions by adsorption/elution with calcium phosphate, aluminum hydroxide gel, or ion exchange resin. We have concentrated Prothrombin Complex by membrane ultrafiltration with no loss of biological activity. Adjustment of salts, etc. by diafiltration has also been demonstrated. A statistical model is presented relating the flux with bulk protein concentration and bulk liquid velocity. With successful membrane ultrafiltration/diafiltration, more efficient purification schemes for isolation of Prothrombin Complex becomes available. Utilizing 106 daltons cut-off membranes, Factor IX activity is demonstrated both in the retentate as well as in the filtrate.
