Cultured Sympathetic Neurons in the Study of Nerve Growth Factor Action

Abstract

The ability to maintain long-term cultures of rat sympathetic neurons in the absence of nonneuronal cells has facilitated a large number of studies dealing with factors important in neuronal growth and development (Patterson, 1978). The work of Mains and Patterson (1973a) demonstrated that the neurons in such cultures exhibited many of the properties of sympathetic neurons in vivo. Furthermore, the time-course of development of neurotransmitter functions in culture paralleled that seen in vivo (Mains and Patterson, 1973b). In the complete absence of nonneuronal influences, the isolated neurons differentiate along the expected adrenergic pathway, showing a developmental increase in the ability to synthesize, store, release, and transport norepinephrine.(Patterson et al., 1975; Burton and Bunge, 1975). The numerous axonal processes elaborated in these neuronal cultures are electrically active, and the cell bodies are sensitive to iontophoretically applied acetylcholine (O’Lague et al., 1975, 1978a).
