Abstract

Radioimmunoassay (RIA) has been used for several years to measure morphine in biological fluids. Recently some doubt has been cast on the validity of the results obtained. Over-estimation of unconjugated drug can occur with antisera having a relatively low % cross-reaction with the main metabolite of morphine, morphine-3-glucuronide [l]. In this study plasma morphine concentrations were measured using two antisera and the results compared to those from HPLC. The plasma samples (25) were from terminally ill patients being treated with oral doses of morphine sulphate (5–30 mg/dose) or diamorphine-hydrochloride (10–60 mg/dose).
