Abstract
The density of field populations of cyst nematodes is estimated by counting the numbers of individuals in samples of soil. This raises two questions, 1) how to take a sample and of what size, and 2) how to arrive at a representative count of the individuals in the sample (extraction method, biotest). Here only the first question is discussed. Once the sample is in the laboratory an accurate count can be made of both the cysts and eggs contained therein. Whatever subsampling is done (investigating part of the field sample after thorough mixing, taking aliquots from egg suspensions) is supposed to result in variances according to binomial or Poisson distributions. This stage of perfection may not have been attained in all laboratories (Müller, 1983), but it certainly is not impossible to approach it. An extensive discussion of laboratory methods is to be found in Seinhorst (1986).
