Hypercontracture of Isolated Adult Rat Heart Myocytes: Multiple Causes and a Common Mechanism

Abstract

The hypercontracture of adult rat ventricular myocytes is an indication of severe and irreversible damage. Normally rectangular and striated myocytes are converted into grossly disorganized round forms as the consequence of an excessive and seemingly uncontrolled shortening of the myofibrils. Under the light microscope, hypercontracture can often be seen to occur after multiple spontaneous cycles of contraction and partial but incomplete relaxation that gradually reduce the overall length of each myocyte. In the final stages, the myofibrils appear to fuse into a nearly amorphous central mass, and the mitochondria and fragments of the sarcoplasmic reticulum are extruded into large superficial blebs of cytosol (see 1 for typical micrographs).
