Abstract

Insertional mutagenesis is a powerful tool for isolating and characterizing genes involved in development because it allows the simultaneous mutating and tagging of a gene. In mammals, two approaches have been successful in inducing insertional mutants: Transgenic animals have been generated by inserting either retroviruses or recombinant DNA into the germ line. Experiments with the first approach exposed mouse embryos to retroviruses at different stages of development (Jaenisch, 1976; Jähner and Jaenisch, 1980; Jaenisch et al., 1981), while experiments with the second approach microinjected mouse zygotes with recombinant DNA (Wagner et al., 1983; Palmiter et al., 1984; Woychik et al., 1985; Mark et al., 1985).
