Abstract

Some of the earliest evidence suggesting a behavioral role for peptides came from studies of pituitary-adrenal function. For example, De Wied and others demonstrated that the behavioral effects produced by removal of the pituitary could be reversed by adrenocorticotropin hormone (ACTH) and other peptides, such as vasopressin (AVP) (De Wied, 1980). Later work established that these peptides could produce profound effects on extinction of learned responses in normal animals even with synthetic analogs reputedly devoid of endocrinological activity. Variously interpreted as actions on attention (ACTH) or on memory consolidation (AVP), these effects were hypothesized to be mediated via some neural substrate. Where this substrate is located and how these pituitary-derived peptides normally interact with it remain largely unknown.
