Regulation of Tissue-Specific Gene Expression in the Mammary Gland: The Role of the Extracellular Matrix

Abstract

Specific interactions between cells and the extracellular matrix (ECM) with which they are in contact play a vital role in tissue organization (1,2). Such interactions are important for directed migration of embryonic cells during development (1,3,4), and for tissue induction during morphogenesis (3,4). It has been established in many model systems, of which the mammary gland is a prime example, that ECM components can support induction and maintenance of tissue-specific function in epithelial cells placed in culture (5,6,7). One important feature of this induction is the modulation of cell shape by interaction with ECM components (5,6,8). In addition to its effects in regulating cell shape and differentiated gene expression, the ECM also exerts an important influence on cell growth and division (10). In transformed cells, progressive loss of differentiated function, altered morphology, and unchecked growth reflect loss of the ability to respond to the external signals that control these functions in normal cells (11).
