Abstract

The neurologic diseases due to human Picornavirus infections are diverse and each may be caused by a number of different viruses from multiple genera. Since the early 1980s, a reawakening of interest in pico-rnaviruses has occured. The relative simplicity of the picornaviral genome along with the development of recombinant DNA technology, enabling the construction of full-length cDNA viral clones in bacterial plasmids which are infectious for mammalian cells, make the human polioviruses particularly useful for studying viral properties, such as attenuation and virulence. The recent atomic resolution of the three-dimensional structures of several picorna-viruses represents a benchmark in animal virus research. This information has provided a quantum leap forward in the understanding of the structure of animal viruses, particularly those possessing icosa-hedral symmetry. The data from nucleotide sequencing and three-dimensional structure determinations is already being applied to animal pathogenesis, particularly in elucidating molecular mechanisms of disease and in the development of safer recombinant PNA engineered poliovirus vaccines.
