Abstract

Many of the metabolic effects of insulin were shown in the previous chapter to be mediated via alterations in the phosphorylation state of the regulatory enzymes involved. In the majority of cases the effect of insulin is to cause the dephosphorylation of target enzymes. Thus, for instance, the key regulatory enzymes of glycogen synthesis (glycogen synthase), glucose oxidation (pyruvate dehydrogenase), lipolysis (hormone-sensitive lipase) are all dephosphorylated in response to insulin. It follows that dephosphorylation of regulatory enzymes is a central mechanism of insulin action.
