Abstract

Site-specific toxin delivery has been achieved by coupling monoclonal antibodies to the A chain subunit of phytotoxins such as ricin and abrin. Such hybrid molecules, termed A chain immunotoxins (A ITs), are now currently prepared according to standardized methods and their biological activities have been studied in detail.The major advantage of A ITs is their low nonspecific toxicity in vivo which allows a safe utilization in human patients (see Chapter 7 for review by Jansen et al. and Chapter 27 for review by Laurent et al.). However, although these conjugates showed stringent specific cytotoxicity, their efficiencies vary greatly from one IT to another. Also, A ITs generally proved to be less active than the original whole toxins towards target cells, suggesting that the role of the B chain not only consists of binding the toxin to the cell surface but also of increasing A chain activity, probably by promoting its transmembrane passage into the cytosol.
