Studies of Phospholipase A2 Activity and Prostaglandin E2 Levels in Rat Stomach Following the Ingestion of Fish Oil

Abstract

It has recently become evident that exogenous and endogenous prostaglandins, mostly of the E series (PGE2) exert cytoprotective properties1 and are effective in the treatment of peptic ulcer disease2. Synthesis of prostaglandins is initiated by the release of arachidonic acid through the action of phospholipase A2 on the 2-position of membrane phospholipids. Such an activity has been described in rat gastric mucosa3,4 and we have recently shown that this enzyme has an sn-2 specificity and is essentially associated with microsomal and plasma membranes5. Prostaglandin synthesis can be affected by ingestion of fish oils which are considered to have beneficial effects on the cardiovascular system6. Fish oils are rich in n-3 polyunsaturated fatty acids (PUFA) which were demonstrated to be efficient in lowering plasma cholesterol, thus preventing atherosclerosis and thrombus formation7,8. However, modification of prostaglandin synthesis in various tissues may lead to non-beneficial effects. For example, menhaden oil fed to mice9 altered the spleen prostaglandin production which in turn may affect the immune function of this organ. In the same way, ingestion of cod liver oil resulted in a reduction in kidney PGE2 generation which may influence blood pressure regulation10.
