Preparation and Analysis of Semen Samples

  • Chapter
Book cover In Vitro Fertilization and Embryo Transfer

Abstract

Washing of spermatozoa to ensure the removal of seminal plasma is an important step in the preparation of the semen sample for in vitro fertilization (IVF). A variety of seminal plasmaassociated factors have been shown to influence sperm function and fertilization, in both positive and detrimental ways (Van der Ven et al., 1982, 1983; Rogers et al., 1983; Sokoloski and Wolf, 1983). Since the seminal plasma constituents of the semen are usually not present (or are tremendously diluted) at the site of fertilization in vivo, it is important that the sperm be washed free from these components prior to insemination in vitro. Since the effects of seminal plasma on sperm may be variable and dependent on the semen source, the semen sample is routinely washed as soon as possible after liquefaction has occurred.
