Synthesis and Properties of Oligonucleotides

Abstract

Developments in the chemical synthesis of polynucleotides have been very rapid. A prediction made in a review article(1) that “advances in the techniques of chemical synthesis of polynucleotides would make nucleic acids more accessible than synthetic polypeptides” seems to have been realized by improvements in the solid-phase synthesis of DNA fragments. A short review, written in 1982, only covers early developments in the syntheses of genes.(2) DNA sequence analyses have been accelerated by recombinant DNA technology and now provide the primary structure of genes in many organisms. These new techniques and information have changed the way biochemists and chemists deal with biological subjects.
