Abstract

The notion that acetylcholine (ACh) might function as a neurotransmitter in the mammalian CNS was mentioned by Dale in his 1937 William Henry Welch Lectures (Dale, 1938). Almost 50 years later, we have some understanding of this function and of the consequences of dysfunction. The function of ACh in the CNS appears to relate to enhancement of neuronal excitability, and it is significant that the first direct demonstration that increased ACh release in cerebral cortex is associated with increased neuronal activity in unanesthetized animals was made in H. H. Jasper’s laboratory (Celesia and Jasper, 1966). The dysfunctions of central cholinergic neurons appear to relate to conditions associated with memory loss, and it is equally significant that Dr. Jasper makes no contribution to the statistics associating these parameters: it is evident that his central cholinergic neurons are fully functional.
