Techniques for Extraction and Group Separation of Bile Acids

Abstract

When bile acids in biological materials are to be analyzed and identified, there is usually a need for extraction and prepurification procedures. Bile and duodenal fluid are exceptions; when the concentration of bile acids is sufficiently high, these materials may be directly applied to the starting line of paper and thinlayer (TLC) chromatograms (see Ref. 1). There is a continuing development in this area [2] also by application of high-performance [3], reversed-phase [4], and overpressured TLC [5]. Direct analysis of the major bile acids by electrophoresis is possible [6, 7], and isotachophoresis in organic solvents is a recent advance [8]. Bile may be directly injected on high-performance liquid chromatography columns (e.g., Ref. 9; see this volume, Chapter 2), but column-switching techniques with automated sample extraction and cleanup are likely to be more widely used.
