Abstract

Recent advances in the study of membrane ion channels have been made through the use of cell disaggregation and single-cell patch-clamp recording techniques. This approach has been responsible for the rapidly advancing knowledge about specific ion-channel proteins and their control processes in numerous tissues. Ion channels have been studied either by isolating a patch of membrane and recording single-channel events or by examination of the aggregate activity of a single type of channel by precisely controlling the ionic species available on both sides of the cell membrane. These techniques have led to the characterization of channels in terms of surface receptor coupling, ion selectivity, voltage activation, voltage-dependent inactivation, and modulation by cellular second messengers. Recent parallel advances in the isolation and sequencing of channel proteins, their reconstitution into lipid bilayers, and the expression of channel proteins in xenopus oocytes by injection of RNA have led to specific hypotheses about their three-dimensional structure, interaction with other membrane proteins, and regulation of synthesis.
