MPTP: Twenty Questions

The last five years have witnessed a remarkable outpouring of scientific literature on 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine or MPTP, a newly described parkinsonogenic neurotoxin. Perhaps the only thing that has surpassed this meteoric rise has been the speed with which concepts regarding its mechanism of action have left the realm of active investigation and gained status as conventional wisdom. From a scientific standpoint, one could argue that this is unfortunate. It will be the thesis of this article that it is time to step back and critically look at what we actually know and don’t know about this compound.
