Genetic Damage in Peripheral Lymphocytes of Chronic Alcoholics

Summary

A possible impact of long-term overexposure to ethanol was studied in two groups of chronic alcoholics in a psychiatric hospital. The hypothesis that acetaldehyde may act as a mutagen for somatic cells and induce free radicals in alcoholics tissues was tested. To monitor the amount of injury resulting from alcohol abuse the following markers were used: chromosome aberrations and unscheduled DNA synthesis (UDS) in peripheral lymphocytes and lipid peroxidation (LPO) in plasma and peripheral lymphocytes. The following results were observed: 2.05% aberrant cells (AB.C) in alcoholics vs. 1.29% in the control group; 12 weeks abstinence decreased the frequency of chromosome aberrations from 2.13 X AB.C to 1.52% AB.C. If the patients were supplemented by 1000 mg of ascorbic acid and 400 mg of α-tocopherol per day in the course of 12 weeks, original UDS T/C increased 3.74 to 4.12 (patients without antioxidants = 3.48, controls = 4.72), LPO in plasma decreased from 1.57 to 1.37 nmol MDA/ml (untreated patients = 1.89, controls = 1.12), LPO in lymphocytes decreased from 1.72 to 1.34 nmol/mg protein (untreated patients = 1.96, controls = 1.14).
