Abstract
Gene targeting, the designed alteration of genomic information by homologous recombination, has provided a powerful means for the genetic analysis of the mammalian systems. The targeted correction of mutations is believed to provide an ideal means for the therapy of diseases caused by mutations. This would be a better alternative, in many respects, to the strategy of expressing wild type c-DNA — complementation approach — in particular, from the point of view of gene expression and the absence of potential mutagenesis.
