The Use of Metal Ions as Spectroscopic Probes in the Elucidation of pKa’s of Small Molecules

Abstract

The apparent pKas’ of the functional groups interacted with metal ions in solutions were determined using a technique devised in this laboratory (Farzami 1992). In these studies several groups of molecules such as coenzyme thiamine diphosphate and some of its derivatives, several dipeptides, amino acid and nucleotides and their bases were the subject of our studies. The results obtained were correlated with that obtained by other techniques such as NMR. In instances where the metal ion could indirectly alter the ionization properties of a group, a conformational scheme could be drawn. This method proved applicable to larger molecules such as proteins, enzymes and nucleic acids (Farzami 1990–1992).
