Abstract

For many years the study of bone disease has been hampered by the technical difficulties of investigating cellular metabolism in calcified tissue. Although this difficulty has not been completely overcome, the development of microradiographic, tetracycline labeling, and calcium autoradiographic methods have made it possible to determine the sites and extent of bone formation and destruction. Investigations in which these methods were used have added significantly to the basic knowledge of bone physiology and to an increased understanding of the pathogenesis of some bone diseases. Since collagen, a basic component of the osseous matrix, has a unique amino acid composition, it is possible to assess bone matrix turnover by measuring the urinary content of hydroxyproline. Measurements of serum alkaline phosphatase also permit an assessment of osteoblastic activity.
