Review of the Structural Studies on Bleomycin

Abstract

More than 12 years have passed since we started our structural study of bleomycin (BLM). These studies are summarized in this chapter. BLM was isolated as a mixture of more than 10 components from the culture filtrate of Streptomyces verticillus. These were separated by CM-Sephadex column chromatography(1) and found to be different from one another in their terminal amine moieties(2). BLMs thus isolated were blue-colored equimolar copper complexes. BLM has not yet been crystallized, so that we could not utilize X-ray crystallographic analysis. In the initial stages of our structural studies, we used exclusively metal-free BLM A2, the main component of the natural BLMs. Metal-free BLM was prepared by treatment of the copper complex with hydrogen sulfide in methanolic solution.
