Metal Complex of Bleomycin and Its Implication for the Mechanism of Bleomycin Action

Abstract

Bleomycin (BLM) is isolated from the fermentation broth of Streptomyces verticillus as a blue powder of its equimolar Cu(II) complex. This copper comes from inorganic cupric salt added to the fermentation medium; without addition of the copper, the fermentation yield of BLM is significantly reduced. The chelated copper can be removed from BLM by precipitation with hydrogen sulfide in methanol solution. Treatment of metal-free BLM with an excess of an inorganic cupric salt, such as cupric chloride, in neutral aqueous solution followed by chromatographic separation on CM-Sephadex regenerates the original equimolar Cu(II) complex in excellent yield, although there are many potential coordination sites in BLM.
