Abstract

DNA damage and repair assays constitute one group of short-term tests that have recently been developed to help assess the carcinogenic and mutagenic potential of chemicals. The alkaline elution assay (AE) can be utilized to measure DNA damage and repair either in vitro or in vivo. This procedure was first described by Kohn et al. (Kohn and Grimek-Ewig, 1973; Kohn et al., 1974, 1976) as a sensitive method for detecting single-strand breaks and alkalilabile sites in DNA of L1210 cells exposed to cancer chemotherapeutic agents. It has been subsequently used to evaluate DNA damage and repair induced in vitro (Brambilla et al., 1978a, 1978b; Cavanna et al, 1977; Fornace and Little, 1979; Swenberg et al, 1976) and in vivo (Brambilla et al., 1978a, 1978b; Eastman and Bresnick, 1979; Parodi et al., 1978; Petzold and Swenberg, 1978) by a wide variety of chemical and environmental agents.
