Abstract

Microbial assays are gaining acceptance as valuable procedures for the recognition of environmental chemicals possessing the potential for inducing cancer. Recent studies have shown that assays based upon the preferential inhibition of DNA repair- deficient bacteria, both as adjuncts to other assays (such as the Salmonella mutagenicity procedure) as well as in their own right, have a high probability of detecting carcinogens (Poirier and deSerres, 1979; Rosenkranz and Poirier, 1979) as DNA-modifying agents. One of the assays which has undergone extensive evaluation and validation is the one based upon the preferential inhibition of DNA polymerase I-deficient E. coli (Slater et al., 1971; Rosenkranz and Leifer, 1980). The present report is concerned with a description of the various modes in which this assay can be run, with the advantages and limitations of each, and includes a comparison of responses in this assay and the potential for inducing cancers in animals.
