A Study of the Edman Degradation in the Assessment of the Purity of Synthetic Peptides

Abstract

The level of amino acid omission in the chain assembly of peptides synthesized by the solid phase method can be determined by quantitating “preview” in the Edman degradation of the peptide, while it is still attached to the resin (Ref. 1). Previous reports indicated a minimum level of 0.3 to 0.5% preview per step(Refs. 2,3). The improved synthetic methodology for solid phase peptide synthesis recently developed in this laboratory (Refs. 4–6) had been shown, by direct analysis of the crude products after cleavage from the resin, to give rise to less than 0.05% amino acid omission per step in the assembly of simple model peptides (Ref. 5).
