Abstract

Much of the literature concerned with clinical disorders of mineral homeostatis contains speculations regarding vitamin D “sensitivity” and vitamin D “resistance.” Whereas vitamin D depletion and excess can be confirmed by bioassay of the antirachitic activity in serum or tissue extracts,1–4 the bioassay techniques are tedious, costly, and imprecise. The introduction of radioactive antirachitic sterols provided new tools that allowed further understanding of the metabolism of these substances.5,6 Since the parent vitamin is now recognized to be metabolized to more potent compounds in the liver and kidney, the lack of specificity of the bioassay of simple serum or tissue extracts is easily understood (Table 1).
