The Use of Enzymatic Radioisotopic Microassays for the Quantification of β-Phenylethylamine, Phenylethanolamine, Tyramine and Octopamine

Abstract

Enzymatic radioisotopic microassays have been used to quantitate those biogenic amines present in mammalian brain in small amounts. Methods were developed for β-phenylethylamine, phenylethanolamine, octopamine and tyramine. The use of partially purified and group-specific N-methyl and 0-methyltransferases, combined with selective procedures for the complete separation of the reaction products, including chromatographic steps, resulted in specific and sensitive microassays. Similar procedures are used for the determination of the activities of enzymes which synthesize or metabolize these amines.

The use of such methods allows the study of levels and metabolism of amines in discrete areas of mammalian brain, plasma, and in single cells from invertebrates, where these amines might play a role as neurotransmitters.
