Abstract
The recent discovery of oncogenes in human and animal tumors has made it possible to investigate the molecular basis of neoplastic transformation. Oncogenes are altered versions of normal cellular genes, generically known as proto-oncogenes (25, 67). Although the role of these proto-oncogenes is for the most part unknown, it is generally believed that they code for proteins functional in the regulation of cell differentiation and/or proliferation. Recent findings demonstrating a high degree of similarity between certain oncogenes and growth factors or growth factor receptors corroborate this general hypothesis. Whereas the sis oncogene has been found to be one of the polypeptide chains of the platelet-derived growth factor, the erb-B oncogene appears to be a truncated form of the epidermal growth factor receptor (18,19,75). Activation of such growth regulatory proteins by mutations, overexpression, or expression in inappropriate cells would be expected to result in uncontrolled growth, the hallmark of transformed cells.
