Cloned HLA-D Genes: Characterisation and Approaches to Expression and Analysis of Function

Abstract

The HLA-D region (Class II) genes determine cell surface glycoproteins which are heterodimers of an α and β chain, and which function in the immune response. This brief paper contains a summary of some of the current studies in this laboratory on the organisation of human Class II genes. Details on the characterisation of six different Class IIα chain genes are presented, including a new gene, DZα, which does not fit into any of the three most clearly established sets of loci: DP,DQ and DR(l). The genes for which we have most sequence information, and the most detailed map, are in the DP region(2). We have succeeded in transferring some of these cloned HLA-DP genes into mouse L cells and can detect expression of DP antigens on the cell surface. The transfectants are apparently capable of inducing a human T lymphocyte clone to proliferate specifically in response to presentation of the neuraminidase glycoprotein of influenza A virus along with DPw2 ClassII determinants(3).
