Abstract

For many years we have been measuring distances between proteins in the 30S ribosomal subunit of Escherichia coli in order to determine their positions in the ribosome. A useful property of this triangulation process is that it permits the construction of increasingly complete and accurate models for the structure as data accumulate. In this respect, distance finding differs strikingly from crystallographic analysis where after a long period of data collection and analysis during which little, if anything, can be said about the structure, one goes from a state of ignorance to full knowledge in a relatively short time.
