Abstract

The mechanism(s) by which professional killer cells lyse target cells remains one of the more important and exciting problems in biology today. Of particular interest to immunologists are killer cells that lyse targets efficiently by a contact-dependent process. This surface contact can be mediated by means of antibodies directed against target surface, in the case of antibody-dependent, cell-mediated cytotoxicity (ADCC) mediated by Fc receptor-bearing cells (macrophages, neutrophils, eosinophils, NK cells) or via receptors (specific and nonspecific) that recognize target surface components in the absence of antibody, as in the case of activated macrophages and cytotoxic T-lymphocytes. In both instances, it is assumed that binding of the target elicits release of the soluble mediators into the diffusion-limited intercellular space. The search for mediators of cytolysis that might explain the cell-mediated cytotoxicity, particularly that mediated by immune effector cells, has resulted in the identification of numerous mediators produced and secreted by these cells. Cytolytic proteins that exert lethal action on targets are presumed to be released into the extracellular space during effector-target cell contact. The increased interest in these effector mediators is largely the result of the feasibility of their isolation in the laboratory in high yields and their further characterization by conventional biochemical and biophysical techniques.
