Abstract
The combination of freeze-fracture and immunocytochemical techniques permits to analyze the distribution of membrane components on both plasma and intracellular membranes (1–3). In particular the fracture-label approach, where cells are directly immunolabeled after freeze-fracture allows the exposition to the immunolabeling of the freeze-fractured inner nuclear membrane. Moreover, since during freeze-fracture, transmembrane proteins partition with the inner (protoplasmic) or outer (exoplasmic) leaflets of the freeze-fractured membrane, the fracture-label approach can also assess this partition. Therefore, we decided here to apply fracture-label technique to EBV producer lymphoblastoid cells in order to investigate the partition on fracture, distribution and extent of glycosylation of EBV envelope glycoproteins during the budding process.
