Abstract

Retroviruses are widely distributed in avian and mammalian species, and cause various neoplastic diseases in their natural hosts (1). The type-C retroviruses have been divided into two groups: the replication-competent viruses that induce leukemia in susceptible animals with a long latent period, but do not usually transform cells in culture; and the acute replication-defective viruses that cause leukemias or sarcomas in animals within a few days or weeks and transform cells in vitro. The replication defect of acutely transforming retroviruses is due to the loss of structural genes that are essential for replication with the concomitant acquisition of altered cellular sequences, named oncogenes, responsible for cell transformation. As a result of their replication defectiveness, acutely transforming retroviruses require a helper replication-competent virus for propagation.
