Abstract

In 1959 when I went to work in Jerard Hurwitz’s laboratory, bacterial enzymes that synthesized DNA and RNA had been discovered. There was good reason to believe that the bacterial DNA polymerase was responsible for the replication of DNA. However, it appeared unlikely that the bacterial enzyme that synthesized RNA, polynucleotide phosphorylase, was responsible for the in vivo synthesis of RNA. A template was not required, and the concentrations of ribonucleoside diphosphates required for synthesis were much higher than present in the cell. (Although no eukaryotic counterpart of the bacterial enzyme had been found, this in itself was not a serious objection).
