Histone and Nucleosome Function in Yeast

Abstract

The nucleosome, which contains two molecules each of histones H2A, H2B, H3, and H4, provides the basis for folding a long, negatively charged DNA polymer into a small nuclear compartment. However, the state of chromosomal condensation is reversible in response to replication, transcription, and chromosomal segregation. How the nucleosome is involved in these functions is a focus of our laboratory. Using reverse genetics we have asked which cellular functions are disturbed when histone proteins are mutated.
