Abstract
Serotonin (5-hydroxytryptamine or 5-HT) is an indole neurotransmitter of the central and peripheral nervous systems. Pharmacological studies of serotonin receptors have revealed considerable heterogeneity of serotonin binding sites; as a result, a variety of receptor subtypes have been designated. Biochemical approaches have also revealed the involve-ment of second messengers in the serotonin-mediated receptor mechanism. Results obtained from autoradiographic methods combined with information from pharmacological studies have allowed the anatomical localization of the serotonin sensitive neurons in the central nervous system. The amino acid sequence of the receptor can be deduced from purified proteins and by molecular biological techniques. Further structural studies and studies of the functional receptor complex, however, require protein preparations isolated from brain tissues. This article will present a method for isolating serotonin receptors by affinity chromatography.
