Mass Spectrometry in Carbohydrate Recognition Studies

Abstract

The neoglycolipid technology has developed into a powerful means of singling out saccharide ligands for carbohydrate-binding proteins. The technology involves the conjugation of oligosaccharides (single components or mixtures) by reductive amination to an aminophospholipid to permit their resolution as neoglycolipids on TLC plates or immobilized on plastic wells for binding studies with soluble and cell-associated carbohydrate-binding proteins. Methods have been developed for high sensitivity LSIMS in situ analysis of resolved neoglycolipid components directly from the silica gel surface. Spectra are obtained from which molecular composition, sequence and branching of oligosaccharides can be determined. The approach is ideal for structure/function assignments and for identification of novel oligosaccharide ligands.
