Human Mycoses: The Role of Molecular Biology

Abstract

Since the early 1980s a dramatic rise in the incidence of invasive disease due to the filamentous fungi (molds) has occurred (Latgé, 1999). Because these fungi are primarily opportunistic pathogens, in large part this trend can be attributed to the increase in the number of patients undergoing highly immunosuppressive therapies including chemotherapy and solid organ and bone marrow transplantation. While the list of organisms infecting immunocompromised hosts is long, the vast majority of these infections are due to Aspergillus species. Among these species, Aspergillus fumigatus accounts for almost 90% of cases (Latgé, 1999). For those infections not due to Aspergillus sp., zygomycetes, Pseudallescheria boydii, and Fusarium sp., are also common pathogens. In this chapter, we will review advances in the study of these organisms as pathogens, particularly the progress made possible by the application of the techniques of molecular biology. We will focus our attention primarily on Aspergillus sp. and the zygomycetes as the most important pathogens in this group. While there has been substantial research on Fusarium sp., it has been predominately on the role of this organism in plant pathogenicity and is beyond the scope of this chapter.
