Abstract

Anthrax has recently gained much attention due to the potential use of Bacillus anthracis spores as an agent of biowarfare. In the event of intentional dissemination of Bacillus anthracis spores, the most likely routes of infection are through the nasopharyngeal and gastrointestinal tracts. Since inhalational and gastrointestinal anthrax are the most severe and potentially lethal forms of the disease, appropriate vaccine strategies are needed to provide optimal protection against these mucosally initiated infections. This chapter will review the role of anthrax toxins in the pathogenesis of anthrax, with special emphasis on how Bacillus anthracis lethal toxin and edema toxin affect immune cells and alter innate and adaptive immunity. We also will summarize traditional strategies for vaccination against anthrax and discuss new approaches to inducing both mucosal and systemic immunity against anthrax toxins for enhanced protection against inhalational and gastrointestinal anthrax.
