Induction of Trifluorothymidine Resistance in Mouse Lymphoma L5178Y Tk+− Cells Following Treatment with Benzidine and 4,4″-Diaminoterphenyl in vitro

Summary

Benzidine (BZD) and 4,4″-diaminoterphenyl (DAT) were tested for their ability to induce resistance to trifluorothymidine in mouse lymphoma L5178Y Tk+/− cells in vitro. BZD induced decreases in cell survival with concomitant increases in the frequency of trifluoro-thymidine-resistant colonies in the presence of S9 mix derived from the livers of rats pretreated with Aroclor 1254, and also in the absence of exogenous metabolic activation. DAT was also cytotoxic and mutagenic in this system, although, in contrast to BZD, its activity was only evident in the presence of the S9 mix.
