Variation of the T-system in different types of intrafusal muscle fibre of cat spindles

Abstract

A modified staining technique by Forbes & Sperelakis (1980) for demonstrating membrane systems of lizard skeletal muscles has been applied to investigate the occurrence of these systems in cat intrafusal muscle fibres. The technique requires a fixative consisting of glutaraldehyde, dextrose and dextran (40,000–90,000MW), all at 3% in an aqueous solution, with 50mM calcium chloride added, pH 7.4. Post-fixation is completed in a solution of 2% osmium tetroxide and 0.8% potassium ferrocyanide in 0.1M sodium cacodylate, then followed by block-staining in a saturated uranyl acetate aqueous solution.
