Abstract
Chronic treatment in clinically relevant doses with typical and atypical antidepressant drugs (desipramine, imipramine, alaproclate and zimelidine) demonstrates marked regional differences in the adaptive changes taking place in 3H-5-HT binding sites, in the 3H-spiperone binding sites linked to 5-HT receptors and in the 3H-imipramine binding sites linked to the 5-HT uptake mechanism. Furthermore, chronic reserpine treatment has also revealed regional differences in the adaptive changes taking place in 3H-imipramine binding sites. These results indicate that chronic antidepressant treatment may cause up-regulation or down-regulation of 5-HT neurotransmission, depending upon the area analyzed. In certain areas like the dorsal cerebral cortex the main outcome of chronic antidepressant treatment may be a stabilization of 5-HT neurotransmission. Therefore, it is difficult to link antidepressant activity of the tested compounds to any generalized change in 5-HT neurotransmission. Furthermore, different antidepressant drugs have been shown to produce different adaptive changes in the 5-HT receptor mechanisms in the same cortical area.
